If you do take a plane, Flamenco Air has the most frequent flights from San Juan and seems to be pretty reliable about leaving on time. We flew Vieques Air Link and while they were very nice and we enjoyed the flight, they only fly in the very early a.m. and late afternoon, and our flight to Culebra was re-routed through Vieques (we switched planes there) because they didn't have enough passengers, apparently. Flamenco seems to be the only airline that primarily flies to and from Culebra from Isla Grande airport.
Flying to Culebra was great and very convenient, but not for the faint of heart! For us it was a highlight of the trip, but man, I'm glad it wasn't too windy the days we flew.
