I believe that is Easter Sunday... Call Carlos jeep rental, but it will be a very hard day for you, since there will be many people there.
That is the worst day to go to Culebra, maybe you will not be able to come back.
You need to get in the ferry line at least 1 1/2 hour before the ticket office opens ( 0800 ) then you will have to get to Culebra ticket office to wait loooong before 0300, if you wish to get back.
My advise... do not go.
